Because in the pre-season they'd be playing against 3rd string or worse defensive players so what's the point? The pre-season at that point is to see who makes the team, not to get in-game practice. In the season they're still playing against the real defense and are playing a "real" in-game situation which will help them improve for later in the year.
I might be willing to give you Peyton if all he was doing was handing the ball off (I wasn't watching), but there's no way benching Eli, or Mathis, is better than giving them more real-game practice all for the sake of a miniscule chance of injury. Like I said, if you want to argue giving the backups in-game practice is more valuable, fine, but benching them to prevent injury just doesn't make sense.
